Potential curability of newly diagnosed acute promyelocytic leukemia without use of chemotherapy: the example of liposomal all-trans retinoic acid.

چکیده
speculated that bHLH SCL functions against WT SCL through the competition for binding to these proteins. To further explore the underlying biochemical mechanisms, we have sequenced some of these coprecipitated proteins, which we hope will be reported in the near future. We disagree with the comment by the questioners that we should be able to show the rescue of the effect of bHLH SCL with WT SCL. This is not an appropriate experiment to show the dominant-negative effect of bHLH SCL. Although the biochemical mechanisms need to be further disclosed, clear are our findings on the distinct biologic functions of WT SCL and bHLH SCL on the commitment fate determination of hematopoietic stem cells. We hope that our ongoing study will give a clear answer to the mechanisms for how bHLH SCL functions in a dominant-negative fashion against WT SCL.

CLINICAL OBSERVATIONS, INTERVENTIONS, AND THERAPEUTIC TRIALS Treatment of newly diagnosed and relapsed acute promyelocytic leukemia with intravenous liposomal all-trans retinoic acid
A novel intravenous liposomal formulation of all-trans retinoic acid (ATRA) was evaluated in 69 patients with acute promyelocytic leukemia (APL): 32 new diagnoses, 35 relapses, and 2 oral ATRA failures. Liposomal ATRA (90 mg/m2) was administered every other day until complete remission (CR) or a maximum of 56 days. Treatment following CR was liposomal ATRA with or without chemotherapy. In an in...

متن کاملThe role of all-trans-retinoic acid in the treatment of acute promyelocytic leukemia.
All-trans-retinoic acid (ATRA) has demonstrated in vivo and in vitro that it could differentiate leukemic cells in acute promyelocytic leukemia (APL). ATRA yields complete remission (CR) rates of approximately 90% in newly diagnosed and first relapsing APL, in APL resistant to one or two courses of chemotherapy, whereas patients in second and subsequent remissions often have only partial remiss...
متن کاملAll-Trans Retinoic Acid plus Arsenic Trioxide versus All-Trans Retinoic Acid plus Chemotherapy for Newly Diagnosed Acute Promyelocytic Leukemia: A Meta-Analysis
BACKGROUND Recently, the all-trans retinoic acid (ATRA) plus arsenic trioxide (ATO) protocol has become a promising first-line therapeutic approach in patients with newly diagnosed acute promyelocytic leukemia (APL), but its benefits compared with standard ATRA plus chemotherapy regimen needs to be proven. Herein, we conducted a meta-analysis comparing the efficacy of ATRA plus ATO with ATRA pl...
متن کاملMolecular basis for the diagnosis and treatment of acute promyelocytic leukemia
Acute promyelocytic leukemia is characterized by gene rearrangements that always involve the retinoic acid receptor alpha on chromosome 15. In the majority of patients t(15;17) is detected, which generates the promyelocytic leukemia gene/retinoic acid receptor alpha rearrangement. This rearrangement interacts with several proteins, including the native promyelocytic leukemia gene, thus causing ...
